    Multi-Stage Modeling of the Kinetics of Activation of CaMKII

    Ca 2+ /calmodulin-dependent protein kinase 2 (CaMKII) plays an important role in induction of long-term potentiation and formation of memory. It is abundant in dendritic spines, and is activated when Ca 2+ flows into the postsynaptic cytosol through open NMDA-type glutamate receptors. Its function is fine-tuned through interaction with other proteins as well as through subunit interactions and regulatory autophosphorylation. We have undertaken a multi-stage project to study the critical kinetics of activation of CaMKII in the spine by combining modeling and experimental studies. We are using computational modeling and simulations on various platforms, coupled with biochemical experiments in vitro, and eventually in vivo, to understand CaMKII regulation. The project includes the following steps: 1. Determining the parameters governing activation of a monomeric subunit. The CaMKII holoenzyme is a large dodecamer of similar, homologous subunits held together by interactions between the association domains located at the carboxyl end of each subunit. Individual, monomeric subunits can be expressed recombinantly by removing the association domain. Computer simulations of activation of monomeric CaMKII by Ca 2+ /calmodulin at both saturating and non-saturating concentrations in a test tube have helped to identify the binding parameters that are most crucial for modeling of regulation of CaMKII and thus have indicated the most useful biochemical assays to measure those parameters (Pepke et al., 2010). We are using these measurements to fine-tune our model of activation of individual catalytic subunits. 2. Building a model of the holoenzyme. Because a CaMKII holoenzyme contains 12 similar subunits, each of which can exist in several states, the holoenzyme can have a large number of state combinations. Thus, modeling the entire holoenzyme requires a computational framework that avoids the ensuing combinatorial complexity. The stochastic simulator MCell provides an elegant, rule-based way of modeling state changes in the CaMKII holoenzyme. 3. Modeling cooperativity that arises from the dodecameric structure of CaMKII. Autophosphorylation at threonine-286, which activates CaMKII subunits, is an inter-subunit event. Thus, it is greatly facilitated by the close proximity of subunits in the holoenzyme. In addition, the subunits within the holoenzyme are arranged as dimers which appears to result in cooperativity in the binding of Ca 2+ /CaM to individual subunits of the dimer (Chao et al., 2010). An accurate model of activation of subunits in the holoenzyme and their autophosphorylation will allow us to explore the effects of cooperativity on CaMKII activation on various time scales. 4. Modeling CaMKII within the context of a postsynaptic spine CaMKII interacts with a variety of other proteins, both in the postsynaptic density (PSD), close to major sources of Ca 2+ influx, and in other parts of the spine. In the fourth stage of this project we plan to implement kinetic models of activation of CaMKII in the context of an MCell model of Ca 2+ influx into a spine upon activation of NMDA-type glutamate receptors (Keller et al., 2008; Keller et al., 2011, submitted).     Manufacture a composite material with a matrix resin epoxy and marble statuary powder as filler is one industry-utilization of waste. Mechanical properties of the composite material is influenced by the particle size of the particles. variations in particle size would form different mechanical properties. This research was conducted to investigate the effect variations in particle size on the mechanical properties of the composite particles. In this research variation in particle size marble statuary used is 60 mesh, 100 mesh, 140 mesh and 200 mesh. Resin used is epoxy resin, and composition of the volume ratio between the matrix and the particles used in research was 80%: 20%. The method is used in the manufacture of composite hand lay-up method. Tests performed in this research is XRD testing on solid marble, bending strength, hardness, and SEM. XRD results obtained from the testing phase identified in the marble statuary are Ca2O5Si , CaCO3, CaO, SiO2 , MgO , MgCO3 , and SiC. Hardness value of composite particles of marble is 22.3 to 48.33 HB, and flexural strength values of composite particles of marble statuary is from 13.89 to 38.89 N/mm . Particle size is good for filler in the composite is 140 mesh particles. Marble composite particles 140 mesh particle size has a flexural strength and hardness highest, because SEM photograph composite of 140 mesh particle distribution in the matrix evenly, and there is a good bond between the matrix and particles of marble statuary

    The Kinematic and Chemical Properties of a Potential Core-Forming Clump: Perseus B1-E

    We present 13CO and C18O (1-0), (2-1), and (3-2) maps towards the core-forming Perseus B1-E clump using observations from the James Clerk Maxwell Telescope (JCMT), Submillimeter Telescope (SMT) of the Arizona Radio Observatory, and IRAM 30 m telescope. We find that the 13CO and C18O line emission both have very complex velocity structures, indicative of multiple velocity components within the ambient gas. The (1-0) transitions reveal a radial velocity gradient across B1-E of 1 km/s/pc that increases from north-west to south-east, whereas the majority of the Perseus cloud has a radial velocity gradient increasing from south-west to north-east. In contrast, we see no evidence of a velocity gradient associated with the denser Herschel-identified substructures in B1-E. Additionally, the denser substructures have much lower systemic motions than the ambient clump material, which indicates that they are likely decoupled from the large-scale gas. Nevertheless, these substructures themselves have broad line widths (0.4 km/s) similar to that of the C18O gas in the clump, which suggests they inherited their kinematic properties from the larger-scale, moderately dense gas. Finally, we find evidence of C18O depletion only toward one substructure, B1-E2, which is also the only object with narrow (transonic) line widths. We suggest that as prestellar cores form, their chemical and kinematic properties are linked in evolution, such that these objects must first dissipate their turbulence before they deplete in CO.Comment: Accepted by ApJ, 34 pages, 12 figure

    Asupan makanan yang melebihi kebutuhan tubuh akan menyebabkan kelebihan berat badan. Sebaliknya, asupan makanan kurang dari yang dibutuhkan akan menyebabkan tubuh menjadi rentan terhadap penyakit. Data hasil Riskesdas tahun 2010 menunjukan status gizi di Sulawesi Utara, sangat pendek 1,4%, pendek 31,8% sangat kurus 0,0%, kurus 3,6%, normal 94,3%, dan gemuk 2,1%. Penelitian ini menggunakan metode observasional analitik dengan pendekatan cross sectional yang dilakukan pada bulan Januari-Maret tahun 2016 pada pelajar di SMP Spektrum Malalayang Kota Manado dan SMP Kristen Lahai Roi Malalayang Kota Manado. Jumlah populasi pada penelitian ini yaitu 94 pelajar. Analisa data menggunakan korelasi spearman. Data yang dikumpulkan menggunakkan metode wawancara, kuesioner food recall 2x24 jam dan pengukuran Antropometri (Tinggi Badan, dan Berat Badan). Hasil penelitian ini menunjukkan bahwa asupan energi yang cukup sebanyak 57 pelajar (60,6%) dan sebanyak 79 pelajar (84%) memiliki status gizi normal. Berdasarkan analisis statistik dengan menggunakan uji korelasi spearman nilai koefisien korelasi sebesar r=-0,388 dan taraf signifikan atau nilai p sebesar 0,000 (p <0,05). Terdapat hubungan antara asupan energi dengan status gizi pada pelajar di SMP Spektrum Manado dan SMP Kristen Lahai Roi Manado. Kata Kunci: Pelajar, Asupan Energi, Status Gizi ABSTRACTFood intake exceeds body requirements will cause overweight. This research use analytic observational method with cross sectional approach conducted in January-March the year 2016 at students in the Junior High School Spectrum Malalayang Manado City and Christian Junior High School Lahai Roi Malalayang Manado City. The number of population in this research totalled 94 students. Data analysis using the spearman correlation. Data collected by questionnaire method interview, use their food recall 2x24 hours and measurements of Anthropometry (height and weight). The results of this study showed that intake of sufficient energy amounted to 57 students (60.6%) and as much as 79 students (84%) had normal nutritional status. Based on statistical analysis using the spearman correlation coefficient test correlation of r =-0.388 and significant extent or value of 0.000 p (p < 0.05).There is a significant correlation between the energy intake and the nutritional status of students in the Junior High School Spectrum Malalayang Manado City and Christian Junior High School Lahai Roi Malalayang Manado City. Keywords: Learner, Energy Intake, Nutritional Status

    Combined antiapoptotic and antioxidant approach to acute neuroprotection for stroke in hypertensive rats

    We hypothesized that targeting key points in the ischemic cascade with combined neuroglobin (Ngb) overexpression and c-jun N-terminal kinase (JNK) inhibition (SP600125) would offer greater neuroprotection than single treatment after in vitro hypoxia/reoxygenation and in a randomized, blinded in vivo experimental stroke study using a clinically relevant rat strain. Male spontaneously hypertensive stroke-prone rats underwent transient middle cerebral artery occlusion (tMCAO) and were divided into the following groups: tMCAO; tMCAO+control GFP-expressing canine adenovirus-2, CAVGFP; tMCAO+Ngb-expressing CAV-2, CAVNgb; tMCAO+SP600125; tMCAO+CAVNgb+SP600125; or sham procedure. Rats were assessed till day 14 for neurologic outcome before infarct determination. In vitro, combined lentivirus-mediated Ngb overexpression+SP600125 significantly reduced oxidative stress and apoptosis compared with single treatment(s) after hypoxia/reoxygenation in B50 cells. In vivo, infarct volume was significantly reduced by CAVNgb, SP600125, and further by CAVNgb+SP600125. The number of Ngb-positive cells in the peri-infarct cortex and striatum was significantly increased 14 days after tMCAO in animals receiving CAVNgb. Neurologic outcome, measured using a 32-point neurologic score, significantly improved with CAVNgb+SP600125 compared with single treatments at 14 days after tMCAO. Combined Ngb overexpression with JNK inhibition reduced hypoxia/reoxygenation-induced oxidative stress and apoptosis in cultured neurons and reduced infarct and improved neurologic outcome more than single therapy after in vivo experimental stroke in hypertensive rats

    Positive postpartum depression screening practices and subsequent mental health treatment for low-income women in Western countries: a systematic literature review

    Problem statement and significance Left undiagnosed and/or untreated, the short-and long-term sequelae of postpartum depression may negatively impact both mother and child. In Western countries, access to mental health care is influenced by socioeconomic factors. The objective of this systematic literature review is to compile factors that hinder and improve access to postpartum depression treatment in low-income women after a positive screen for postpartum depression. The key question of focus is: what are the characteristics associated with access to mental health treatment for low-income women with a positive postpartum depression screen in Western countries? Methods A PRISMA-based systematic literature review was conducted of studies published in English before February 2016 that looked at treatment for postpartum depression in low-income women who had been identified with the condition. PubMed and EBSCO databases were searched using MESH and key terms and found 100 articles that met the selection criteria. After review by two independent researchers, 18 studies with 17 unique populations were included in the literature review. Results Two independent abstractors searched the included articles for themes surrounding impediments and advantages for low-income women identified with postpartum depression in obtaining mental health treatment. Characteristics of successful mental health treatment included studies that employed the use of a home visitor and those that separated outcomes for women with previous mental health treatment. Themes that emerged as treatment obstacles included cultural barriers, physical barriers, systemic health care barriers, and social barriers. Implications for practice This review will help to better inform screening and treatment priorities for those in the medical field who may encounter women experiencing postpartum depression and are not aware of the various barriers to care specific to low-income women. This review will also help policymakers identify specific obstacles that are not addressed in postpartum screening mandate policies which can affect the implementation of these policies
